Mouse intracerebral (i.c.) toxicity studies with 29 pyrethroids confirmed earlier mouse i.p. and rat oral and i.v. findings in 3 respects: .alpha.-cyano-3-phenoxybenzyl esters produced choreoathetosis, convulsions and salivation, while compounds lacking the .alpha.-cyano group yielded tremors and convulsions; high stereospecificity was involved in inducing both poisoning syndromes; large toxicity differences for (1R,trans) vs. (1R,cis) resmethrin and permethrin did not extend to ethanomethrin and the cyanophenoxybenzyl esters. I.c. investigations established that: profuse salivation was not unique for pyrethroids with the .alpha.-cyano group; the inactivity of (1R,trans) resmethrin in the brain was not due to detoxification; pyrethroids acting most rapidly in the brain were those with the highest knockdown activity for insects; the cyanophenoxybenzyl esters, in comparsion with the non-cyano pyrethroids, had a high i.c. toxicity relative to their synergized i.p. toxicity indicating the importance of the brain in the type 2 poisoning syndrome.
