10 rat dams and their pups were included in the study (121 rats). Breeder rats and newborn rats were housed with other rats known to harbourP. carinii carinii(Pcc). Rat pups were sampled by the oral swab/polymerase chain reaction technique at ages 1-4 weeks (8 pups at 1 week, 13 pups at 2 weeks and 11 pups at 4 weeks). All 31 samples and their dams (n=4) were positive for Pcc-specific DNA by. A second set of oral swans were collected from 3 rat dams and their pups at 1-2 h, 24 h and 1 week after birth. 78% (28 of 36) of the pups were positive for Pcc-specific DNA within the first 2 h of life. By 24 h and at 1 week of age, 100% were positive for Pcc-specific DNA. The 3 dams were also positive for Pcc-specific DNA during all sample time points. Rat pups were collected by caesarean section at 3 time points (2-4 weeks of gestation). None of the 48 rat fetuses, placentas or deciduae were positive for Pcc-specific DNA. The 3 dams were positive by oral swab, lung tissue homogenate and spleen homogenate.
