Under laboratory conditions, 36 sequential subinoculations of a single strain of Plasmodium vivax malaria during which suboptimal treatments were employed gave rise to a significant increase in trophozoite resistance to the drug primaquine. The maximum dose of primaquine tolerated by human volunteers no longer had any effect on the parasitemia developed by the last few subinoculations. Apparently fully-viable gametocytes could be transferred to mosquitoes and develop into sporo-zoites, although such sporozoites failed to infect man. The capacity of primaquine to cause the development of a resistant P. vivax strain of malarial organisms indicates the desirability of using a good blood schizontocide in addition to primaquine therapy. || ABSTRACT AUTHORS: A C.
