Techniques developed for isolating milk oligosaccharides since the earlier report [Methods in Enzymology 28, 24] and structures of newly found sugars, are reviewed. Bio-Gel P-4 columns have been found superior to Sephadex G-25 columns in gel permeation chromatography, and borate paper electrophoresis has been found useful for the separation of oligosaccharides with Fuc alpha 1 'right arrow' 2Gal grouping from isomeric oligosaccharide mixtures. Isolation of the following oligosaccharides from human milk is described: 6'-galactosyllactose; lacto-N-fucopentaose; difucosyl-p-lacto-N-hexaose and difucosyl-p-lacto-N-neohexaose; and fucosyllacto-N-octaose and fucosyllacto-N-neooctaose.
