The order of 17 L-arabinose mutants of Escherichia coli B/r has been determined by 3-and 4-factor transduction experiments with phage Plbt. The assortment of the markers threonine and leucine, located on either side of the arabinose cluster, was observed among recombinants between pairs of arabinose mutants. The sites of the arabinose mutants are arranged in a linear sequence. The arabinose mutants are divided into 3 functional groups. Group A (4 mutants) accumulates trace amounts of a keto sugar and is inhibited by L-arabinose; Group B (8 mutants) accumulates large quantities of the keto sugar, ribulose, and is inhibited by L-arabinose; Group C (5 mutants) accumulates small amounts of a keto sugar (more than Group A) and is resistant to L-arabinose. Group C mutants are distinguished from mutants in Groups A and B by abortive transduction tests. Comparison of the order of the mutant sites with the grouping by functional criteria demonstrates complete correspondence between the order of the mutants and the groups to which they belong. The transduction experiments involving pairs of arabinose mutants were characterized by the presence of marked negative interference.
