In all types of mammalian striated muscle the structure of the contractile system is remarkably similar, suggesting that the mechanism of the transduction process varies very little in the different forms of this tissue. On the other hand, the time course of the response to stimulation is one of the ways in which the various kinds of muscle differ from each other. Indeed the pattern of this response is used to distinguish the two types of cell of which skeletal muscle is composed, theslow and fast fibres. As it is now generally agreed that contraction is a consequence of the hydrolysis of ATP, the manner in which the ATPase activity of the myofibril is regulated will be a most important factor in determining the nature of the physiological response. The contractile responses characteristic for different types of muscle will therefore depend largely on the differences in the mechanisms present for regulation of the myofibrillar ATPase. A number of factors determine the way in which myofibrillar ATPase is regulated. These include the type of innervation of the muscle, the properties of the Ca2+-transport
