INTRODUCTION The magnesium ion is probably the most under-appreciated of all of the biologically significant ions. While most researchers are familiar with the role of Mg in stabilizing DNA and membranes and as a required cofactor for the normal function of many Mg-dependent enzymes [1,2], they often do not give thought to whether they should include Mg in their experimental solutions or whether experimental manipulations may affect available Mg. Few consider the notion that intracellular levels might be regulated or that they could be subject to change in a physiologically relevant manner. In this review, we examine current ideas about the role of Mg in excitable cells and discuss the latest information about the way in which cells regulate their internal Mg levels.
