Observation of the first stages of infection of cells by the flagellates of Leishmania donovani was carried out using tissues from both hamster and mouse origin raised in a culture medium of a mixture of horse and rabbit sera plus some yeast extract and Hank's fluid. Only monocytes were readily entered. The parasite penetrated the cell with the flagellum trailing. Once inside the cells, the Leishmania then moved about freely with the flagellum in the lead. Both the monocyte and the contained parasites break down quickly, but the parasites usually break down and are partially digested before the phagocyte breaks up. As they die, the Leishmania bodies become vacuolated and a small drop-let appears at the end of the flagellum. || ABSTRACT AUTHORS: A. C. Walton
