Synthesis of complement components C2, C3, C4 and C5 by rheumatoid synovial tissue was demonstrated by three independent technics: in vivo metabolic studies with Radio-labeled C3; in vitro biosynthesis of C2, C3, C4 and C5; and staining of cells in rheumatoid synovium with fluorescent antiserums to C3 and C4. Production of complement components was more frequently observed with cultures of tissue obtained from patients with rheumatoid arthritis than with those from patients with degenerative or traumatic arthritis. The findings demonstrate the importance of local production of components of the inflammatory response in rheumatoid arthritis. (N Engl J Med 290:1284–1288, 1974)
