The lymph nodes of rats were cultured in vitro and the radiosensitivity of the lymphocytes was compared with that of similar cells in vivo. Cell counts from film prepns. gave the percentage of lymphocytes which were pyknotic 5 hrs. after various doses of X-rays. From the dose-effect curve, the ED50 (dose to produce 50% pyknosis in 5 hrs.) was 275 r in vitro and 150 r in vivo. The high radiosensitivity of lymph-node lymphocytes is probably a property of the cells themselves, while the indirect effects of radiation are slight or nil. Medium lymphocytes were about as sensitive as small lymphocytes. Reticular cells, monocytes and plasma cells were unaffected by the doses used. Whole blood was cultured and irradiated in vitro and the ED50 for blood lymphocytes was 1600 r. Pyknotic lymphocytes were found in the peripheral blood of rats after 400 r whole-body irradiation, but they were rapidly removed from the blood stream by the lungs. The scattered lymphocytes in the intestinal villi were relatively resistant, the ED50 being 2250 r.
