The results of studies with perfused adrenals,'taken together with analysis of urine from normal individuals, 2 as well as those suffering from Cushing's di~ ease,~ identified cortisol and corticosterone as the predominant secretory products of the adrenal gland. Subsequently, an enzyme capable of oxidizing cortisol to cortisone was reported in rat liver. 4 Given the name 1 lp-HSD, it was found to reversibly convert cortisol and corticosterone to their inactive 1 1-oxygenated pairs, cortisone and ll-dehydrocorticosterone. 5 The presence of l lp-HSD has now been detected in diverse tissues. Although 11p-HSD activity has been localized primarily in the microsomes of liver: kidney, l placenta,'and~ pleen,~ significant activity is also reported in nuclei of kidneylo and brain." Moreover, the similarity in K, values for kidney microsomal and nuclear 11P-HSD activityI0 suggests that the enzymes are identical or similar. Surprisingly, reported values of K, and pH that are optimal for 1 la-HSD activity vary over an unusually wide range from tissue to tissue and they vary even more widely from laboratory to laboratory for the same tissue. Monder and WhiteI2 listed variability in substrate and cofactor concentrations, pH, and temperature of incubation to account for this lack of reproducibility and gave special emphasis to differences in methods of tissue preparations among investigators.
