Considerable attention has been given to the possible involve ment of steroid hormones in psychiatric disorders, and the gluco corticoid receptor is of interest as a potential target for abnor malities underlying these conditions. A recent report of a trisomy of the 5ql 1.2—* q 13.3 region translocated to chromosome I seg regating with schizophrenia in an uncle-nephew pair (Bassett et al.. 1988). together with somatic cell hybrid mapping of the glucocor ticoid receptor (GRL) locus to this same region (Weinberger et al., 1985) strengthened the hypothesis of ctiologic involvement of GRL in schizophrenia. Thus, linkage studies between schizo phrenia and markers in this area, including GRL. became an ob vious research strategy.In order to localize GRL among other RFLP markers, we con ducted linkage analyses of several chromosome 5 loci in three large kindreds. Our data indicate that GRL is not located in proximal 5q. but is closely linked to D5S22 which has been mapped near the end of the long arm of chromosome 5 (Leppert et al.. 1987).
