This review summarizes recent considerations germane to the mecha nism(s) for NaCI absorption by the mammalian proximal nephron. Most arguments concerning epithelial NaCI transport derive from the hypotheses formulated by H. H. Ussing and his associates in their analysis of Na+ fluxes in anuran epithelia. Their evocative model derives from three major consid erations. Ussing (69) and Ussing & Zerahn (71) demonstrated that net Na+ transport was identical to the zero-voltage short-circuit current and occurred against an electrochemical gradient. Then Koefoed-Johnsen & Ussing (33) proposed that Na+ entered cells passively at apical surfaces and was actively transported out of cells at basolateral surfaces via an active Na+ /K+ exchange pump. Finally, Ussing & Windhager (70) provided strong evidence that dissipative ion transport in epithelia might involve a paracellular route. The application of Ussing's paradigm to the mamm alian proximal renal tubule also requires analysis of certain other factors. Of particular impor tance in this context are the following:
