Method.All the instruments and glassware used in the experiments' were sterilized; the instruments were sterilized in a flame continually during the preparation of the cultures, so that infection of the cultures was rare. The solution was warmed and poured into small Petri dishes. The heart was aseptically removed from the chick embryo in Locke-Lewis solution and transferred to another Petri dish containing about 10 co. of the medium. Here it was cut into small pieces and transferred by means of a pipette to a cover: glass, which was immediately inverted over a hard vaseline ring on a depression slide. The chance of evaporation by this method of the Lewises is very small, as the tissue can be placed on the cover-glass and sealed in a few seconds. When the tissue was to be planted in hypertonic or hypotonic solution the heart was placed in the liquid immediately after its removal from the embryo; the tissue was therefore never in contact with other solutions that might possibly dilute the medium.
