ObjectiveTo investigate the osteogenic effect of KLD-12 polypeptide/recombinant human bone morphogenetic protein 2 (rhBMP-2) fiber gel composite scaffolds seeded with rabbit bone marrow mesenchymal stem cells (BMSCs) that were implanted in rabbit gluteal muscle.MethodsBMSCs were isolated and cultured by a density gradient method, and a muscle bag model was established in the gluteal muscle of rabbits. Each graft (experimental group A: KLD-12 polypeptide/rhBMP-2/BMSC gel; positive control group B: KLD-12 polypeptide/rhBMP-2 gel; positive control group C: rhBMP-2; negative control group D: KLD-12 gel) was separately implanted into the muscle pocket of the gluteal muscle. The rabbits were sacrificed at 4, 8, and 12 weeks postoperation for X-ray analysis, gross observation, micro-CT analysis, histomorphological staining, and an alkaline phosphatase (ALP) activity assay.ResultsX-ray, gross observation and micro-CT results showed that osteogenesis among groups A, B and C was the most obvious in group A, while group D showed no obvious osteogenesis. Hematoxylin-eosin (HE) staining, Giemsa staining and ALP activity results showed that mature bone tissue was formed in groups A, B and C and that mature bone tissue formation was the most obvious in group A.ConclusionThe KLD-12 polypeptide/rhBMP-2 gel seeded with BMSCs showed good osteogenesis in the gluteal muscle. BMSCs and rhBMP-2, which were located within the KLD-12 polypeptide gel, exerted a synergistic osteogenic effect.
