A rapid and sensitive assay for chitinolytic activity has been developed on a micro-scale. This assay is based on the use of fluorogenic 4-methylumbelliferone glycosides of N-acetylglucosamine oligosaccharides in order to characterise the enzymes associated with chitinolytic microorganisms, i.e. exo- and endochitinases and N-acetylglucosamimidase. The sensitivity of the assay allows detailed kinetic studies to be performed. Examples of its uses are given for chitinolytic activities from an estuarine bacterial isolate.
