5-HT 3 receptors in the periphery mediate depolarizing effects of 5-HT on neural tissue (see Richardson and Buchheit, 1988). Recently 5-HT 3 binding sites have been found in the brain (Kilpatrick et al., 1987). No certain functional correlates for these binding sites have been identified although behavioral effects of selective 5-HT 3 antagonists have been described (eg, Costall et al., 1987). We now report that activation of the 5-HT 3 receptor markedly increases the release of dopamine (DA) from rat striatal slices. Striatal slices (400/~ m thick) from eight-week old, male Sprague-Dawley rats were superfused in a thermostatic chamber (37 C, 300/~ 1 volume), at a rate of 0.5 ml/min with artificial cerebrospinal fluid (aCSF; see figure legend for composition) containing tyrosine (50/~ M), nomifensine (10/~ M) to block DA reuptake, and pargyline (10/~ M) to block monoamine oxidase, aCSF was continously gassed with 95% 02 and 5% CO 2. Sixty minutes after the start of superfusion, 1.5 ml fractions were collected into tubes containing 166# 1 of a solution of 1 N perchloric acid, 10/~ M ascorbic acid and 2.5 pg epinine (internal standard). Slices were exposed to the substances under investigation by transferring the chamber inflow tubing to reservoirs containing aCSF of appropriate composition. Samples were kept on ice during the collection period and stored at-20 C until assayed.
