"Superovulation" in rats induced by treatment with pregnant mare serum gonadotropin (PMS) and human chorionic gonadotropin (HCG) was analyzed by measuring numbers and size of follicles, and number of ova, in normal and hypophysectomized rats given these hormones. In normal rats and in rats treated with HCG during estrus, follicles of 55,000,000 cubic mcm or larger ovulate spontaneously. 5-50 IU PMS accelerated development of increased numbers of large follicals to 20-60 per rat, but injection of 15-45 IU HCG at 2 pm, 1-4 days later permitted few ovulations and left half the follicles retained. 5, 10, or 50 IU PMS (no HCG) evoked spontaneous ovulation 3 or 4 days later (5 days after estrus is normal). These rats given 10 or 50 IU PMS had cystic follicles with luteinized walls and entrapped ova, on the day after ovulation. Rats given 10-35 IU PMS in estrus did not ovulate, and those given 50-80 IU PMS released only a few ova. As an estimate of the amount of endogenous LH released, rats were hypophysectomized at various times and given various doses of HCG. Results of hypophysectomy experiments suggested that endogenous LH is sufficient after 5 or 50-80 IU PMS, but is subnormal after 10-35 IU.
