CONSIDERABLE recent evidence indicates that the secretion of noradrenaline (NA) from sympathetic nerves, on arrival of the propagated nerve impulse, is not an all-or-none but a graded and controlled physiological process. One control mechanism appears to be mediated by the local formation of prostaglandin E (PGE)1. Thus sympathetic nerve stimulation causes formation and release of large amounts of PGE2,3. Addition of exogenous PGE at concentrations known to occur physiologically severely depresses sympathetic neurotransmitter secretion4–6. Blocking local formation of PGE enhances nerve stimulation induced secretion of NA in several tissues and species7–10, suggesting removal of a PGE-mediated negative feed-back control of NA secretion.
