METHODSThe animals were anesthetized (Nembutal), and the left labyrinth was surgically exposed under aseptic conditions. A single branch of the vestibular nerve that innervated a specific receptor crista or macula was isolated and cut immediately adjacent to the receptor neuroepithelium. Free crystals of either horseradish peroxidase (HRP) or a horseradish peroxidase/cholera-toxin mixture (HRP-CTP) were applied to the cut nerve stump. Next, bone wax was applied over the area, the wound sutured closed, and analgesic (Butorphanol) administered. Following a survival period of 48 to 96 hours, the animals were given an overdose of anesthetic and perfused transcardially with heparinized saline and aldehyde fixatives. Frozen serial sections were cut in either the transverse or parasagittal planes, then processed histochemically using either diamino benzidine (DAB) or tetramethyl benzidine (TMB) as a chromagen for the HRP and HRP-CTP tracing tissue. Afferent fibers and their terminal distributions were visualized and photographed using light microscopy, then drawn for comparison using either camera lucida and a digipad or direct video camera image tracing on a computer screen with reconstruction software. For delineation of possible tracer spread between the intended cut nerve branch and other vestibular organs, the receptor neuroepithelia were examined in several animals, with no evidence of tracer spread being observed.
