The last minisymposium in this series on the topic of the prothymocyte [1] and the subsequent summary [2] came at the beginning of a time of explosive growth in cell and molecular biological approaches to all stages of T cell ontogeny. However, despite these advances, we, and our colleagues who have written the papers herewith, would all agree not only that the major questions of the earliest stages of T cell ontogeny remain unresolved, but also that the complexity of the questions has increased alongside the information. Ultimately this will surely lead to greater understanding of pre-T and early T cell development.It is appropriate, first, to restate the original concept of the'prothymocyte'. This supposedly was an extrathymic cell (as distinguished from an intrathymic cell type) which was not known to bear surface markers characteristic of cells within the thymus but which could repopulate an irradiated thymus. It presumably also fulfilled such a function in the primary development of the thymus during embryogenesis [1-3].
