An improved method for spore membrane isolation was developed based on sucrose density gradient centrifugation in a vertical rotor. The advantage of this over previous methods was the complete removal of RNA and a 40% reduction in protein content, while retaining the high specific activities for membrane bound dehydrogenases and for amino acid uptake.
