SUMMARY IL-2production byperipheral blood mononuclear cells (PBM)isdecreased inpatients withsystemic lupuserythematosus (SLE). Thisdefect can bereversed bytheremoval ofCD8+lymphocytes. The purposeofthese studies wastodetermine whether theCD8+IL-2 suppressorcells comprise aspecific subset orwhether all CD8+cells havethis activity. Lymphocyte subsets wereidentified andseparated bytwo-colour flow cytometry prior toa48hmitogen stimulation. TheCD8+cells that suppressed IL-2production co-expressed HLA DR andwere radiosensitive. Othermarkers co-expressed by CD8+cells which arefound on suppressorcells such asLeu15(CDI1),Leu11 (CD16),andLeu7 werealso found ontheCD8+IL-2 suppressorcell population inSLE.Inhealthy subjects, removal of CD16+,butnotofCD8+cells markedly elevated theproduction ofIL-2. TheCD8-CD16+non-T cell subset suppressed IL-2production bynormalandSLEPBM inautologous andallogeneic combinations. Thissubset may beahumanequivalent ofthemurine natural suppressorcells. These results demonstrate thatthecells that suppressIL-2production inSLEareheterogeneous, and suggest thattheybelong tomore than one lineage.
