Bioassay of androgen relies on the increase in weight of seminal vesicle and prostate gland in a castrated rat. Eisenberg and Gordan(1950)used the levator ani method for the measurement of protein anabolic effect. These methods all utilize the direct action of androgen on each target organ atrophied by castration. The most ideal method for bioassay in general is to carry out experiments under the most sensitive physiological condition. Adrenal excretes androgen and estrogen, together with corticoids which have important bearing on the metabolism. Consequently, it was considered that the sensitivity would be more increased by castration and removal of the adrenal at the same time than only by castration and,if this were true,results somewhat different from adult rats would be obtained with immature rats.Reaction to androgen was therefore examined in adult and immature rats,castrated or castrated-adrenalectomized,in order to examine the effect of adrenal on the bioassay of androgen.Further,examinations were also made on the sensitivity of various organs to androgen and some new observations were gained.
