In the mammalian cholinergic neuron, acetylcholine (ACh) and ACh-esterase (AChE) are transported in an anterograde direction in the non-terminal axon at fast rates (about 150 mmld and 250400 mmld, respectively, see Haggendal et al. 1971, Lubinska & Niemierko 1971, Ranish & Ochs 1972, Dahlstrom et al. 1974). The synthesizing enzyme, choline acetyltransferase (CAT), however, is transported at much slower rates (cf. Saunders et al. 1973, TuEek 1975), in accordance with its cytoplasmic localization (cf. Fonnum 1970). The'transportable'fraction of ACh in the axon probably is localized within an organelle which can protect the accumulated ACh from the huge amounts of accumulated AChE, bound to the sER membranes (cf. Kasa et al. 1973). In the nerve terminal endings, ACh is considered to be stored in the small clear synaptic vesicles, found densely packed both in motor end plates and other cholinergic nerve endings (cf. review by Whittaker 1969). The ACh storing organelle in the axon is, however, not identified, although the large dense cored vesicles, observed proximal to a crush in cholinergic nerves (Woog & Bennet 1972), have been suggested to contain ACh (Dahlstrom et al.
