N-Nitrosomorpholine Synthesis in Rodents Exposed to Nitrogen Dioxide and Morpholine

We examined the possibility thatN-nitrosomorpholine (NMOR) was producedin vivowhen mice were gavaged with morpholine and exposed to atmospheric nitrogen dioxide (NO2). Using a "stopping solution" adjusted to pH 1 that contained ascorbate and sulfamate to prevent artifactual nitrosation, andcis-2,6-dimethylmorpho1ine to detect any such nitrosation, we demonstrated that no significant NMOR was formed in rat stomach contents or blood, or the whole mouse, after exposure to morpholine and NO2. We verified the results of Iqbalet al. (Science207:1475, 1980) that NMOR was produced when mice were treated with morpholine and NO2, and the whole mouse homogenate in aqueous methanol was worked up without using a stopping solution. We showed that the NMOR was produced in the homogenate by a nitrosating agent present in several organs of NO2-exposed mice.
