In cultured cells, permeability can be altered by treatment with cytoskeleton disrupting agents or chemically generated toxic oxygen metabolites. In epithelial cells, which form tight junctions in culture, disruption of microfilaments by cytochalasin B resulted in opening of tight junctions and a decrease in electrical resistance (9). Cytochalasin B also fragmented microfilaments and increased permeability to albumin in endothelial cell monolayers (10). A similar disruption of actin filaments and a concomitant increase in perme-
