Geminiviruses are a group of circular, single-stranded DNA viruses that cause a number of significant diseases of cash and subsistence crops in tropical and subtropical areas (1). Recently, the incidence of diseases caused by geminiviruses has been increasing in more temperate environments, including southern regions of Europe and the United States (2, 3). Dicotinfecting geminiviruses have either monopartite or bipartite (designated genomic components DNA A and DNA B) genomes and are transmitted by either leafhoppers or whiteflies (4).Primers for the amplification of approximately half the DNA A genomic components of whitefly-transmitted geminiviruses have been reported (5). We report the sequences of a pair of degenerate primers suitable for the PCR-mediated amplification of near full-length genomes (or DNA A genomic components) of geminiviruses that infect dicotyledonous plants. The products of these amplifications (spanning all but 17 bp of the genome) are useful not only as diagnostic probes but also for the determination of DNA sequence and single-cutting restriction endonuclease sites prior to cloning the viruses.
