Drosophila homeobox-containing genes have been shown to regulate the expression of one another as well as other classes of development-regulating genes at the genetic and molecular le~ el.~,~~ For example, the levels of expression of Antp and Ultrabithorax (Ubx) in the primordia of parasegments 2, 4, and 6 are dependent upon the proper expression of jkhi tarazu (ftz). 26 An intriguing cross-regulatory interaction is exhibited byfrz and even-skipped (eve), in which the absence offtz expression in some neurons results in the elimination of eve expression. 27 Direct evidence for cross-regulation is also inferred from the several recent studies in Drosophila using in vitro binding assays and in vivo co-transfection assays. 6J7. 25 Nothing is known about a potential cross-regulatory function of mammalian homeobox-containing genes. Since one would predict that cross-regulation via transcription factors would have to occur in a cell-autonomous manner, it will be important to ascertain the cellular site of expression of specific homeobox-containing genes. Spermatogenesis offers an excellent system in which to address this question in the mammal because, as is discussed below, it is possible to define the precise cell types expressing homeobox-containing genes.
