A dose-related hypotension occurred after oral administration of minoxidil to unanesthetized rats, dogs and monkeys. The duration of the hypotensive effect after a single dose usually exceeded 24 hours. The hypotensive effect was accompanied by a marked increase in cardiac output and rate. Daily administration to renal hypertensive dogs caused a lowering of blood pressure wihich was sustained throughout a treatment period of three weeks. Acute experiments in anesthetized dogs demonstrated that minoxidil exerted a relaxant effect on vascular smooth muscle, which could not be prevented by beta adrenergic, cholinergic or histaminergic blocking agents. Minoxidil reduced sympathetic vasoconstrictor responses in both anesthetized and unanesthetized dogs without reducing the response to injected norepinephrine. It is concluded that the hypotensive effect of this agent is due primarily to a vasodilator action, although attenuation of sympathetic vasoconstrictor nerve function may also contribute. Minoxidil exerts minimal pharmacologic effects on systems other than the cardiovascular system and is nontoxic to rats, monkeys and minipigs at dose levels far in excess of those required for hypotensive activity. In dogs, minoxidil is tolerated at high doses, but a degenerative lesion confined to the right atrium consistently occurred after repetitive administration.
