Changes in DNA synthesis and DNA content of liver cells during liver tumorigenesis by diethyinitrosamine (DENA) in the medaka (Oryzias latipes) was measured by autoradiography and Feulgen-DNA cytofluorometry. The fish were kept for 6 weeks in water containing DENA (100 ppm). A single pulse of 1 μCl tritiated thymidine was given Ip to each fish 2 hours before the fish was killed, and the labeling indices of 5–7 fish were counted throughout the 13 weeks of the experiment After 1 week, the indices of both parenchymal and nonparenchymal cells were at the control level but increased rapidly at 2 weeks and stayed high during the DENA treatment However, the indices began to decrease immediately after the animals were transferred into normal water, Except for the tumor nodules that maintained a high level of labeling frequency, the indices fell to the control level in the later period of the experiment, Furthermore, in parallel with histologic studies, the nuclear DNA content of each hepatocyte was measured by Feulgen-DNA cytofluorometry simulteneously combined with autoradiography. The hepatocytes whose DNA contents were measured might have included normal ones as well as the hepatocytes altered by DENA. DNA contents became widely distributed even before appreciable histologic changes could be detected. During the treatment of the fish with DENA, the DNA contents of labeled hepatocytes ranged from the normal diploid value (2C) to more than 6C. When the tumor nodules were observed histologically, the DNA distribution histogram was found to be similar to the control profile except for the presence of hepatocytes with higher DNA contents around 6C.
