Proflavin, ethidium, and ellipticine, compounds known or thought to bind to nucleic acid helices by intercalation, were found to inhibit the processing of 45 s nucleolar RNA to other RNA species in the ribosomal RNA maturation sequence in L1210 mouse lymphoma cells. The effect is prompt, is exerted on normally formed 45 s nucleolar RNA, and is not secondary to inhibition of RNA or protein synthesis; hence it appears to be due to direct action on nucleolar RNA processing. These findings suggest the possible existence of helical basepaired segments at specific processing sites in 45 s nucleolar RNA.
