Urinary NT-methylhistidine (NMH) excretion has been used as an index of myofibrillar protein degradation (MPD). This paper describes a method for assessment of urinary NMH levels in growing rats of variable thyroid status. Samples urine were collected from control, thyroidectomized (Tx), and triiodothyronine-supplemented thyroidectomized rats (T3) for development of this method. Urinary NMH and standard NMH were deacetylated then derivatized with phenylisothiocyanate (PITC) and injected into a high-performance liquid chromatography (HPLC) system and detected at 254 nm. A peak in urine corresponded in retention time to standard PITC-NMH. Mass spectroscopy of this peak demonstrated that it consisted of a compound of molecular mass equal to that of PITC-NMH. Quantities of NMH excreted by rats in this study were similar to NMH excretion patterns reported in other studies. Thyroidectomy, which reduces MPD, reduced urinary NMH levels whereas T3 administration, which has been reported to enhance MPD, increased urinary NMH excretion. Phenylisothiocyanate derivatization, therefore, provides a rapid and sensitive method for assessing NMH excretion patterns in growing rats.
