A current theory is that butyrate protects against colon cancer by inhibiting colonic cell proliferation and promoting differentiation and perhaps apoptosis. An extension of this theory is that dietary fibre may protect against colon cancer through the production of butyrate by the colonic microflora. Support for these concepts comes from several sources, but an attempt to bring the various lines of reasoning together in one cohesive pattern reveals a number of inconsistencies. The purpose of this review is to clarify the chain of evidence that results in the conclusion that butyrate is protective against colon cancer and that fibre is protective through the production of butyrate. Data will be presented that support or fail to support each step in this reasoning process and inconsistencies will be noted where they exist.
