CHEMICAL CONSTITUENTS OF Stellaria dichotoma var. lanceolata AND THEIR ANTI-INFLAMMATORY EFFECT ON LIPOPOLYSACCHARIDE-STIMULATED RAW 264.7 CELLS

Stellaria dichotoma var. lanceolata Bunge (Radix Stellariae), distributed mainly in Ningxia and neighboring provinces of China, belongs to the genus Stellaria L. The roots of this plant (Stellariae Radix) are used as traditional Chinese medicine (Yin-Chai-Hu) for the treatment of asthenic fever in the late stage of febrile diseases [1]. Previous phytochemical investigations reported that a series of β-carbolines [1, 2], cyclic peptides [3–5], neolignans, and phenylpropanoids [6] have been isolated from the roots of this plant. They show anti-inflammatory [7], anti-allergic [6], and blood vessel dilated effects [5]. Here, we describe the isolation and structural identification of 20 compounds (1–20) and evaluate the anti-inflammatory effects of 11 compounds from Stellaria dichotoma var. lanceolata on lipopolysaccharide (LPS)-stimulated RAW 264.7 cells [8]. From a 70% ethanol extract of Radix Stellariae, 20 compounds (1–20) were obtained. Their structures were characterized as dibutyl phthalate (1), stellarine B (2), trans-2-methylhex-3-ene (3), Z-octadecyl caffeate (4), dichotomide I (5), bis (2-ethylhexyl) phthalate (6), N-tetratriacont-20, 23-dienoic acid (7), 22-dihydrochondrillasterol (8), phthalic acid isodibutyl ester (9), 2-methyl butyl propyl phthalate (10), geleboline C (11), dichotomide XIV (12), 9, 10-bis (2-ethylhexyloxycarbonyl)-N-(1-hexylheptyl)-perylene-3, 4-dicarboximide (13), isovanillic acid (14), bis (2-methylheptyl) phthalate (15), hypoxanthine-9-β-D-ribofuranoside (16), marinacarboline D (17), 1-methyl-α-D-glucose (18), menthalactone (19), and dichotomide XII (20). All the constituents, compounds 3, 4, 6–8, 10, 11, 13, 14, 16, and 18, were isolated for the first time from the genus Stellaria L.
