Methods are suggested for obtaining oocytes at meiotic stages later than diplotene, which is usually reached just before or shortly after birth. One method which has proved successful in some species (e.g., mouse, rat and hamster) is to liberate the oocytes into a culture medium by puncturing the follicles, after which many oocytes immediately resume their maturation. It appears that an inhibitory influence within the follicle rather than a factor in the oocyte causes the dictyate stage to persist, and rupture of the follicle releases the oocyte from the inhibitor. Primate and dog oocytes have not responded so well to this technique, and possible alternatives are indicated.
