The availability of gamma globulin in a form suitable for intravenous use fi. v. Ig) has resulted in its application for treatment of an increasingly large number of infectious and autoimmune diseases. Only recently have some of the specific mechanisms of action of gamma globulin been elucidated. Mechanisms of action can be broadly classified as either neutralization of a specific toxin or microbe, or as immunomodulation. Historically, gamma globulins were first used to enhance opsonization or neutralization of bacteria, viruses or toxins. More recently immunomodulatory functions of gamma globulins have been described which include alterations in cytokine production (Shimozato et al. 1990), inhibition of T-cell proliferation in vitro (Antel et al. 1981), decreases in T-cell activation in vivo (Leung et al. 1987), inhibition of antibody secretion in vitro (Stohl 1986), increases in natural-killer cell activity (Finberg et al. 1992) and increases in antibodydependent cell cytotoxicity (Finberg et al. 1992).
