Two experiments examined the relationship between impairments in electrophysiological measures of hippocampal synaptic plasticity and deficits in working memory in aged animals using the 8-arm spatial maze task. Speed of hippocampal kindling via perforant path stimulation was used as a measure of neuronal plasticity. In Exp I, 3-and 26-mo-old male Fischer 344 rats received electrical stimulation of the ipsilateral dentate gyrus and on the cortical surface bilaterally. Results show a striking relationship between performance in the 8-arm spatial maze and number of trials to reach the kindling criterion for both young and old Ss. Exp II tested the effects of pentoxifylline, choline, pentoxifylline plus choline, and saline on the age-dependent decline in spatial memory in 3-and 26-mo-old male Fischer 344 rats. It was found that the pentoxifylline-treated aged Ss had improved memory and augmented synaptic transmission.
