A CLOSE structural similarity between human growth hormone (HGH) and human placental lactogen (HPL), also known as human chorionic somatomammotropin (HCS), was postulated several years ago on the basis of physiological and immunological studies1–3 and confirmed by the demonstration of a high degree of homology between the two hormones at the amino-terminus, eleven of the first seventeen residues being identical4. Other studies5, 6 indicated similarities in the amino-acid composition of many of the tryptic peptides derived from HGH and HPL, and established homologies in sequence for a small region at the carboxyl terminus. I have determined much of the remaining amino-acid sequence of the HPL molecule. A full description of this work is in preparation. A portion of the amino-terminal sequence of HPL is shown in Fig. 1, together with corresponding regions of HGH7 and ovine prolactin8. Comparison of the sequences of HPL (residues 1–35) and ovine prolactin (residues 10–44) provides good evidence of homology: residues in twelve of the thirty-five positions are identical and a further ten contain pairs of amino-acids related through highly acceptable point mutations in the codon9.
