The removal of 95 per cent of the pancreas in male rats will result in a permanent diabetic state characterized by constant glycosuria and hyperglycemia in 47 per cent of the animals at three months following the operation; at six months 98 per cent of the animals exhibit these symptoms. This permanent diabetes is preceded by a period of incipient diabetes distinguished by abnormal glucose tolerance test and postprandial hyperglycemia and glycosuria. Prior to the onset of the incipient phase, there is a oneto threemonth period of time which has been termed "prediabetic" by Houssay, since the rats are normoglycemic, aglycosuric, and have a normal rate of growth. This prolonged "prediabetic" phase provides the opportunity to study the sequence of changes occurring in the remaining pancreas, and to investigate the factors which cause the development of the overt diabetic syndrome. The purpose of the present study was to determine and correlate the changes in the blood sugar, the serum insulin-like activity (ILA), the insulin content of the pancreas, and the cytology of the islets during the prediabetic and early diabetic periods in partially pancreatectomized rats. MATERIALS AND METHODS Wistar albino male rats, initially weighing ninety to 100 gm. were used. The animals were individually caged and weighed weekly. Under Nembutal anesthesia, eighty-two rats were submitted to a subtotal pancreatectomy according to the technic described by Foglia, which consists of the complete removal of the splenic and gastric portion of the pancreas, and removal of the duodenal portion except that which lies between the common bile duct and the proximal duodenum; this residual tissue will be referred to as the remaining pancreas and represents approximately 5 per cent of the total mass of the organ. The removed tissue was weighed routinely. In fourteen control animals a sham operation was performed by delivering
