Recent interest in the biosynthesis of the chondroitin sulphates has emphasized the need for a procedure permitting the isolation of the constituent monosaccharides in appreciable yields for subsequent analyses etc. The hexuronic acids in particular pose problems since they are extensively destroyed during the strong hydrolysis conditions necessary to release other monosaccharides 1. Enzymic hydrolysis is not practicable 2. Reduction with borohydride of the carboxyl group of the uronic acid residue prior to hydrolysis produces polysaccharide material from which hexose derived from the uronic acid can be obtained in high yield3, 4. However, the transformation of nronic acid to hexose could introduce complications in biosynthetic investigations where [z4Clglucose was being used as a precursor since hexose (mainly galactose) is present in samples of chondroitin sulphate and chondromucoproteinS-L Also small quantities of precursor glucose of high specific activity could be a contaminant. Mild but effective conditions for hydrolysis have been achieved with sulphonated polystyrene resin in dilute acid, eg 0.05 M HC1 (refs. 8-1o). ANASTASSIADIS AND COMMON 10-1~ in particular utilized this technique to hydrolyse carbohydrate-containing materials from animal tissues. A hexosamine-containing fraction was separated from the remainder of the hydrolysis products but no further examination or purification of the fractions was attempted.
