We have now heard back from the three referees who agreed to evaluate your manuscript. As you will see from the reports below, the referees find the topic of your study of potential interest. They, however, raise substantial concerns, which, I am afraid to say, preclude its publication in its present form. In general, the reviewers recognized that this work provides a large and potentially useful dataset relevant to interactions among proteins expressed in the human liver. Nonetheless, they raise a series of issues which they felt would need to be addressed convincingly before this work would be appropriate for publications. I highlight here two issues of particular importance: 1. The reviewers had some important technical concerns regarding the assessment of false-positives and false-negatives from the yeast 2-hybrid screen, as well as concerns regarding the controls used in the co-IP and GST-affinity experiments. See, in particular, the points raised by reviewer #3. Additional clarification, and in some cases additional experimental controls, appear needed. 2. The reviewers felt that the initial selection of the bait proteins had not been sufficiently described, and they worried that the bias introduced by this targeted set of liver-expressed proteins needed to
