With fall in pH below 7 the characteristic absorption maximum of fluorescein at 490 m[mu], is rapidly reduced and disappears completely at pH 2. At pH 7.6 solutions of fluorescein have an absorption maximum at 490 m[mu] with minor bands at 280 and 325 m[mu]. Successful conjugation of globulin to the fluorescein nucleus is accomplished by combination with fluorescein isocyanate which is prepared from fluorescein amine II. The latter has been characterized by its infra red spectrum and a comparison made with the standard sample from the laboratory of Prof. A. H. Coons (Harvard Med. School). The UV absorption spectrum of fluorescein amine II and fluorescein isocyanate in dioxane acetone solution has been obtained, and the absorption spectra of fluorescent globulin conjugates given.
