The author urges a more critical approach to the interpretation of electron-micrographs, and closer collaboration between cytochemists and electron-microscopists. He considers that there should be scepticism about structure revealed at the sub-microscopic level, unless it can be checked independently. For instance, one may make use of the known tendency of particular substances to arrange themselves in particular ways. Thus, phospholipids have a strong tendency to present themselves in the form of parallel or concentric layers. These layers may already have been present in life, but the possibility must be borne in mind that they originally formed part of lipoprotein complexes, were separated off by the "unmasking" effect of the fixative, and only then adopted a lamellar arrangement.
