111In-oxinate and 111In-acetylacetonate (111In-acac) are very widely used compounds for labelling blood cells for clinical and biological studies (Thakur et al, 1977; Danpure & Osman, 1981). However, they both have the drawback that, to obtain high labelling efficiency, the cells must be washed free of plasma before labelling as, in the presence of plasma, indium avidly binds to transferrin. This paper reports an advance in cell labelling with the use of a new water-soluble metal chelate 111In-tropolonate. In contrast to 111In-oxinate and 111In-acac, 111In-tropolonate efficiently labels blood cells in plasma. As a result the cell damage associated with the removal of the plasma is avoided and the time for cell labelling is reduced. A new method of isolating granulocytes is also given.
