The developmental regulation of the contractile proteins of the muscle sarco mere is well orchestrated. The proteins appear in concert during differentia tion and accumulate to stoichiometric levels, and their induction can be examined both in vivo and in vitro. These properties make myofibrillogenesis an ideal model for the examination of coordinate gene expression during a complex ontogenetic event. In recent years, cDNAs and genes encoding many of the contractile proteins have been isolated from a variety of species. This has permitted direct examination of the structural elements that are important for the developmental regulation of these genes and has revealed the contribu tions of both transcriptional and posttranscriptional processes to their expres sion. In addition, such studies have provided insights about trans-acting factors that may mediate the coordinate expression of muscle-specific genes. The knowledge gained from the cloning of contractile protein genes is the focus of this review. Because of space limitations, we concentrate on the study of mammalian contractile protein genes, but we also consider some particularly relevant studies of contractile protein genes from other vertebrate speCIes.
