The physiological role of chroma& vesicles in adrenal medullary cells has been elucidated through studies of the isolated chromaffin vesicles and through studies of evoked secretion employing intact animals, isolated perfused glands, and adrenal medullary cell cultures. I4 The chromafin vesicles contain high concentrations of catecholamines and ATP and they contain the enzyme dopamine/.?-hydroxylase, which catalyzes the oxidation of dopamine to norepinephrine. They are directly involved in the exocytotic secretion of epinephrine and norepinephrine, and the vesicle membranes are equipped with an ATP-dependent transport system for translocating catechol-amines into the vesicles against a high concentration gradient. Although the individual reactions in the formation of norepinephrine and epinephrine have been well characterized, there was only little information on the integrated function of this pathway at the time the studies reported here were undertaken. Isolated chromatlin vesicles can take up ATP, but little was known about the accumulation of ATP or its turnover in chroma& vesicles of intact cells. Studies in which we have examined the flux of catecholamines through chroma& vesicles5 and ATP metabolism in cultures of intact bovine adrenal medullary cells6.'are reported here.
