We have demonstrated that adenovirus type-2 (AD-2) will productively infect cultured human keratinocytes, an ectodermally derived cell. During infection cytopathic effects are observed in suprabasal cells while most basal cells appear to be free of virus. Experiments with cultures of basal cells alone and with cells infected in suspension indicate that this lack of response of the basal cells is not a result of resistance to infection. Apparently the productive response is blocked in basal keratinocytes, but after some unknown maturational events ensue fully permissive and mature infectious particles are produced. Immunofluorescence analysis suggests that blockage of viral expression in the basal cells occurs prior to T-antigen synthesis. These results imply that expression of Ad-2 is dependent upon the state of cellular differentiation in keratinocytes. This system should prove to be a valuable tool in the study of differentiation as well as the nature of latent infection.
