The localization of Walker 256 tumor cells introduced into the peritoneal cavity was studied by transmission electron microscopy, scanning electron microscopy, and autoradiography in rats with intact peritoneum and in rats in which the peritoneum was denuded of mesothelial cells. Implantation into the parietal peritoneum was a rare and late event if the peritoneum was intact. Peritoneum denuded of mesothelial cells showed implantation of tumor cells in less than 2 hr, and the confluence of a layer of tumor cells was frequently observed by 24 hr.
