It is shown that proteolytic enzymes reduce the electrophoretic mobilities of human and chimpanzee erythrocytes, the decreases depending upon the specificity of the particular enzyme. In the case of human erythrocytes, a semiquantitative correlation has been found between these reductions and the quantities of sialic acids released in bound form asN-acylated neuraminic acid-containing mucoids. In contrast, the action of proteolytic enzymes on other animal erythrocytes produces either a small decrease, no change, or in the case of chicken red cells an increase in the electrophoretic mobility.Treatment of animal red cells with neuraminidase produces in all instances a marked reduction in the electrokinetic charge. The sialic acid present on the surface of chicken erythrocytes has been shown to be predominantlyN-acetylneuraminic acid while that from horse is mainlyN-glycolylneuraminic acid.Successive enzyme treatments with neuraminidase, papain, and trypsin reveal marked differences between the erythrocytes from various species.
